The mission of the Berlin Institute of Health at Charité (BIH) is medical translation: transferring biomedical research findings into novel approaches to personalized prediction, prevention, diagnostics and therapies and, conversely, using clinical observations to develop new research ideas. The aim is to deliver relevant medical benefits to patients and the population at large. Since 2021 the BIH has been integrated into Charité as its so-called third pillar.
For the group Prof. Dr. Ludovic Vallier starting from the earliest possible date limited until 31.12.2029 we are looking for a Post Doctoral Fellow (d/f/m) in full-time (38,5h/week).
The Vallier Lab based at the BCRT will takes advantage of human induced pluripotent stem cells and primary organoids to understand liver development in human and to generate cells with a clinical interest for modelling diseases and for cell-based therapies applications. More precisely, the Vallier Lab investigates the molecular mechanisms controlling cell fate decisions during human development and exploits the resulting knowledge to produce liver cells, especially hepatocytes and cholangiocytes.
What you can expect
- The candidate will join the ERC project FunChol which aims to uncover new mechanisms driving chronic liver disease and to identify targets for therapeutic development.
- The role of the candidate will be to investigate the mechanisms by which acquisition of cellular plasticity during liver regeneration can lead to tumorigenesis.
- For that you will derive, characterize and differentiate organoids from the liver of patient with chronic disease and perform genome editing using CRISPR/Cas9.
- The resulting cell lines will be then characterized for their transcriptome, epigenetic state and somatic mutation signature using whole genome analyses.
- Results obtained in vitro will be validated using fresh tissue samples and in vivo models for liver injury.
What you bring along
- PhD or equivalent in stem cell biology and oncology.
- Strong experience in tissue culture, especially with organoids derived from primary tissue, differentiation, and characterization.
- Solid background in cancer biology and gastroenterology.
- Experience with animal models for gastroenterology cancer.
- Experience in flow cytometry, QPCR, expression profiling, confocal imaging, CRISPR/Cas9 genome editing, and ChIP-Seq.
- Experience in working independently while keeping a strong team spirit.
- At least one publication with first authorship.
- Strong self-motivation, organizational skills, and ability to lead and develop scientific projects.
