Working at Charité
The Neumann Research Group is a young, translational research group at the Charité Comprehensive Cancer Center. Our research focuses on patient-derived tumor models, which we use to investigate mechanisms of therapeutic resistance and develop novel treatment strategies for patients with pancreatic ductal adenocarcinoma (PDAC).
PDAC is one of the cancers associated with the poorest prognosis. At the same time, the emergence of novel direct KRAS inhibitors offers, for the first time, the opportunity to therapeutically target a key oncogenic driver of pancreatic cancer. The rapid development of these compounds and their increasing evaluation in clinical trials have made understanding why tumors fail to respond to KRAS inhibitors, or respond only temporarily, a highly relevant challenge in translational cancer research.
In our research group, we combine clinically relevant questions with advanced preclinical tumor models. Our approaches include patient-derived organoids, functional drug testing, and genetic, transcriptomic, and proteomic analyses. A particular focus of this German Cancer Aid (Deutsche Krebshilfe)-funded project is to elucidate KRAS-dependent mechanisms of drug resistance in PDAC. To this end, we have access to an established biobank comprising 36 patient-derived PDAC organoid models representing, among others, different KRAS mutations. These models are used to investigate both intrinsic and acquired resistance to KRAS inhibitors.
The project is conducted in close scientific collaboration with Prof. Dr. Reinhold Schäfer from the Institute of Pathology at Charité. As a co-applicant on the project funded by German Cancer Aid, he contributes his expertise particularly in the molecular and pathological characterization and functional investigation of tumor models. This collaboration brings together the Neumann Research Group's expertise in patient-derived tumor models and functional drug testing with the complementary expertise of the Institute of Pathology.
Overview of the position
You will work as a Research Associate on a research project funded by German Cancer Aid (Deutsche Krebshilfe), focusing on elucidating KRAS-dependent mechanisms of drug resistance in pancreatic ductal adenocarcinoma (PDAC). As part of your Ph.D. research project in the natural sciences, you will investigate the molecular mechanisms underlying intrinsic and acquired resistance to KRAS inhibitors using an established biobank of 36 patient-derived PDAC organoid models.
Your key responsibilities will include:
- Cultivation and functional characterization of patient-derived PDAC organoids
- Completion and analysis of the molecular genetic and transcriptomic characterization of the organoid models
- Conducting and evaluating functional drug sensitivity assays using KRAS inhibitors and established cancer therapies
- Establishment and characterization of organoid models with acquired resistance to KRAS inhibitors
- Identification of potential resistance mechanisms, particularly those involving the KRAS/MAPK signaling pathway and relevant transcription factors
- Functional validation of candidate genes and resistance factors using overexpression, RNA interference, and/or genome editing techniques
- Bioinformatic analysis and integration of genetic, transcriptomic, and functional data
- Investigation of the impact of the tumor microenvironment on therapeutic resistance through co-culture experiments involving PDAC organoids and cancer-associated fibroblasts (CAFs)
- Investigation and functional validation of potential combination therapies, including combinations of KRAS inhibitors with established chemotherapy regimens
- Presentation of research findings at scientific conferences and contribution to scientific publications
- Close collaboration with clinical and scientific research partners as well as Charité's Core Facilities
This position offers the opportunity for further scientific qualification leading to a Ph.D. in the natural sciences. You will work in an interdisciplinary, translational research environment, addressing a clinically highly relevant research question. The overarching goal is to gain a deeper understanding of resistance mechanisms to KRAS inhibitors and to develop novel therapeutic combination strategies for pancreatic cancer.
What we are looking for
You enjoy experimental research, are interested in innovative approaches to translational cancer research, and are eager to contribute to the development of novel therapeutic strategies for pancreatic cancer. You are motivated to combine experimental and computational approaches, ideally have some initial hands-on research experience, and are keen to further develop your scientific and methodological skills as part of your doctoral studies.
We are therefore looking for a highly motivated and dedicated individual who meets the following requirements:
- A completed university degree (e.g. Master's degree, German Diplom, or State Examination) in Biology, Biochemistry, Molecular Biology, Biomedicine, or a related field
- Strong interest in cancer genetics and molecular biology, particularly in mechanisms of therapeutic resistance in pancreatic cancer
- Interest in experimental cell and molecular biology, ideally with initial hands-on experience in cell culture and/or organoid culture
- Interest in the functional investigation of genetic alterations and their impact on treatment response and the development of therapeutic resistance
- Basic knowledge of R, Python, Shell, or comparable programming languages, or a willingness to further develop these skills for the analysis of biological data
- Motivation to integrate experimental research with computational and bioinformatic analyses
- A critical, independent, and structured approach to scientific work, combined with the ambition to independently develop and pursue research questions
- Enthusiasm for interdisciplinary collaboration and scientific exchange with clinical and research partners
- Excellent written and spoken English skills
- Strong motivation to pursue further scientific qualifications and successfully complete a Ph.D.
